  • Biography

    Jonathan V. Sweedler received his Ph.D. in Chemistry from the University of Arizona in 1988, spent several years at Stanford before moving to the University of Illinois at Urbana-Champaign in 1991 where he has been ever since. At Illinois, he is currently the James R. Eiszner Family Endowed Chair in Chemistry, the Director of the School of Chemical Sciences, and has appointments in Neuroscience, Molecular and Integrative Physiology, Bioengineering and Medicine. His research interests focus on developing new approaches for assaying small volume samples, and in applying these methods to study novel interactions between cells. These analytical approaches include capillary separations, micro and nanofluidics, miniaturized separations, NMR, and mass spectrometry. He has used these tools to characterize small molecules and peptides in a range of animal models across the metazoan and in samples as small as individual cells and cellular domains. Sweedler has published more than 500 manuscripts and presented 500 invited lectures. He is currently the Editor-in-Chief for Analytical Chemistry.

    Jonathan Sweedler's research emphasizes two major thrusts. The first is the development and implementation of new analytical instrumentation and methodology related to metabolomics and peptidomics with much of our efforts directed toward scaling these methods to nanoliter and smaller volume levels. We are currently developing a range of mass spectrometry imaging approaches that allow thousands of individual cells to be characterized for their neuropeptide content, and a unique capillary electrophoresis approach that allows us to sample the cytoplasm for a selected neuron or glia and characterize its metabolome. Many of these measurement capabilities are unique and not currently available elsewhere.

    The second area involves characterizing cell to cell signaling across the metazoan; specific queries address what molecules are present in specific cells and networks, and how they change based on network activity, animal behavior, or even on exposure to drugs. Neuropeptides are perhaps the most diverse category of neuromodulators, and the Sweedler group has characterized the neuropeptides and prohormones in the sea slug, honey bee, urchin, planarian, songbird, and in several mammals. Literally hundreds of new prohormones and even more putative neuropeptides have been discovered, and the bioactivity of several of these novel neuropeptides characterized.  They collaborate with multiple groups at the BI and throughout the broader scientific community. 

    Sweedler's major sources of research funding are the NSF, NIH, DOE, and private foundations. Sweedler has been the PI for the past 15 years of the Neuroproteomics and Neurometabolomics of Cell to Cell Signaling, a P30 Center from NIDA.

  • Research

    Jonathan Sweedler's research emphasizes the development and implementation of new analytical instrumentation and methodology, and the elucidation of the functional roles of multiple neurotransmitters and neuromodulatory compounds in individual neurons and well defined neuronal networks in the common neuronal model system, Aplysia californica. Much of his research group's work involves scaling a variety of methods to the nanoliter volume regime to allow the identification and quantitation of neuroactive compounds from cellular microenvironments.

    In terms of analytical methodology (primarily BSG research), Sweedler is developing new microseparation methods based on capillary electrophoresis and chromatography, and has demonstrated unique detection modes such as wavelength resolved fluorescence, radiochemical, and NMR. His group, in collaboration with other members of the Biological Sensors Group, improved the sensitivity so that high quality NMR spectra can be reliably obtained on picomole samples. One of the most significant methods under development in his laboratory involves profiling the peptides in individual neurons using matrix-assisted laser desorption/ionization (MALDI) time-of-flight mass spectrometry.

    A large subset of his group's research (primarily within the NeuroTech Group) is designed to understand the molecular (chemical) nature of learning and behavior. Using the techniques described above, they are studying the neurotransmitters and neuromodulatory compounds used in simple and well-defined invertebrate neuronal networks, such as the bag cell neurons that control aspects of reproduction and several groups of neurons involved with feeding in Aplysia californica. They have discovered many cases of previously unknown processing of known neuropeptides, as well as previously unknown peptides. Increasing our understanding of the functional roles of cotransmission in relatively simple neuronal systems directly contributes to the Biological Intelligence research theme at the Beckman Institute.
